一次運(yùn)行多種pH范圍的IPG膠條，獲得大限度的分離效果，數(shù)據(jù)重現(xiàn)性高

PROTEAN i12 IEF系統(tǒng)可以在一次運(yùn)行中，使用不同pH范圍的IPG膠條，從而獲得大限度的分離效果。選擇pH3-10的寬范圍膠條，比較其與pH3-6，pH5-8，pH7-1三種窄范圍膠條重疊所獲得的2D圖，四種膠條運(yùn)行，一次便可獲得所有數(shù)據(jù)。紅圈內(nèi)突出顯示了窄范圍膠條的高分辨率，表格內(nèi)顯示了三條重復(fù)膠條內(nèi)檢測(cè)到的蛋白點(diǎn)數(shù)，說(shuō)明重復(fù)性非常好。

來(lái)自小鼠肝臟的樣品通過(guò)尿素-硫尿-CHAPS溶液提取。提取液分別用12條不同pH范圍，長(zhǎng)度均為11 cm的ReadyStripTM IPG膠條進(jìn)行等電聚焦試驗(yàn)。所有的膠條都放置在一臺(tái)PROTEAN i12 IEF儀器的聚焦盤內(nèi)，運(yùn)行。IPG膠條的pH范圍分別為：3-10，3-6，5-8和7-10，每種pH范圍重復(fù)3次。第二向SDS-PAGE試驗(yàn)使用濃度為8-16%的梯度CriterionTM預(yù)制膠，用考馬斯藍(lán)染色。上圖為使用不同pH范圍的lPG膠條后獲得的2-D電泳圖和三個(gè)重復(fù)樣本所獲得的蛋白點(diǎn)數(shù)。表格顯示三個(gè)重復(fù)膠條中都存在的蛋白點(diǎn)數(shù)。
